ABSTRACT

Ras is best known for its ability to regulate cell growth, proliferation and differentiation. Mutations in Ras are associated with the abnormal cell proliferation which can result in incidence of all human cancers. Extracellular signal-regulated kinase (ERK) is a downstream effector of Ras and plays important roles in prognosis of tumors. Recently, evidence has gradually accumulated to demonstrate that there are other effectors between Ras and ERK, these proteins interact each other and constitute the thorough Ras/Raf/MEK/ERK signaling pathway. The pathway has profound effects on incidence of esophageal carcinoma and clinical applications of some chemotherapeutic drugs targeting the pathway. Further understanding of the relevant molecular mechanisms of Ras/Raf/MEK/ERK signaling pathway can be helpful for the development of efficient targeting therapeutic approaches which contribute to the treatment of esophageal cancer. In this article, roles of Ras/Raf/MEK/ERK signaling pathway in esophageal carcinoma as well as pharmacological targeting point in the pathway are reviewed.

ABSTRACT

ObjectiveTo study the application of three-dimensional simulated surgical technique in precise hepatectomy. MethodsFrom July 2009 to February 2010, 16 patients with primary liver cancer underwent preoperative simulated imaging and three-dimensional simulation of liver resection.The 3D extent of simulated hepatectomy and actual hepatectomy was compared and analyzed. ResultsThe shape and the extent of the liver resected were very similar in the simulated and the actual hepatectomies. The mean differences in the length, breadth and depth of the remnant livers were 0. 6118 cm,0. 4490 cm and 0. 3199 cm, respectively. ConclusionsSimulation hepatectomy could predict the extent of the actual liver resection, and provided accurate guidance and preoperative planning for precise hepatectomy.

ABSTRACT

Golgi protein-73 (GP73) is a type-lⅡ Golgi glycoprotein localized on the membrane of the Golgi complex.Recently,a growing number of studies have demonstrated that the expression of GP73 is closely correlated with primary hepatocarcinoma (PHC).GP73 has a higher sensitivity and specificity than alpha-fetoprotein (AFP) and it may be a novel serum marker for the detection of PHC,especially for early PHC.This article reviews the relationship between GP73 and PHC.

ABSTRACT

AIM: To investigate the role of mild heat stress towards the immunological function of splenic macrophages and significance of BiP protein expression. METHODS: Primary cultured splenic macrophages were prepared and placed in 41 ℃ thermostat for thermal stress and restored to 37 ℃ after an hour. Heat stressed macrophages were separated into groups at time points of 0 min, 30 min, 60 min, 120 min and 180 min, and their BiP mRNA and BiP protein expressions were detected. At the same time, the phagocytic function, cytotoxicity and chemotaxis of the macrophages were detected. RESULTS: After heat stress, the expressions of BiP mRNA and BiP protein in splenic macrophages remarkably increased, and reached to peak after 30-60 min, still remained higher than control group after 120 min and restored to normal level after 180 min. At the same time, mild heat stress enhanced the phagocytotic, cytotoxicity and chemotactic activities of splenic macrophages, and reached to peak after 60 min then gradually decreased. CONCLUSION: The expression of BiP protein is enhanced after mild heat stress, synchronous changes happen both in BiP protein expression and cellular function. There is close relation between BiP protein expression and increased functions of splenic macrophages induced by mild heat stress.

ABSTRACT

Objective To study the influences by a Cyelin-dependent kinase inhibitor Roscovitine on cell cycle in mitotic hepatoma carcinoma cell SMMC-7721. Methods Microscope,MTT, flow cytometry and R-T PCR were used to observe the effects of Roscovitine on morphology, proliferation, cell cycle, apoptosis and the mRNA expression of CDK2, Caspase-3, bcl-2 in SMMC-7721 cells. Results Roscovitine inhibited the proliferation of SMMC-7721 cells in dosage and time dependent manner and induced apoptosis. Flow cytometry showed the ratio of G0, G1 increased. R-T PCR showed that the expression of bcl-2 reduced, Caspase-3 increased. Conclusion Reseovitine can inhibit the growth, proliferation, block the cell cycle at G0/G1 and promotes apoptosis of mitotic SMMC-7721 cells, and the mechanism of apoptosis is dependent on the activity of bcl-2 and Caspase-3.

In recent years, it is discovered that tissue factor can discriminate and regulate cell signal transduction, promote neogenesis of blood vessel and inflammatory reaction, regulate cell adhesion and movement, have a intimate relationship with invasion and metastasis of malignant tumor. ABSTRACT

OBJECTIVE:To study the curative effect and mechanism of ulinastatin (UTI) administered by two different routes for severe acute pancreatitis (SAP). METHODS: 50 healthy male SD rats were randomly divided into 5 groups: sham operation (SO) group (fed with water ad libitum), SAP model group, positive control(nafamostat) group, ulinastatin peripheral venous perfusion group, and ulinastatin celiac arterial perfusion group. SAP model was established in the latter four groups, and then the rats were treated with corresponding drugs. 12h later, sample tissues were taken from pancreas and lungs for measurement of MPO level and histopathologic observation and the blood samples were collected for measurement of serum amylase, TNF-? and TXB2. RESULTS: Levels of amylase, TNF-?, TXB2, MPO and Hughes histopathologic scores of pancreas and lungs in all drug-treated groups were significantly lower than in SAP group (P

ABSTRACT

<p><b>OBJECTIVE</b>To assess the effect of cholesterol in bile on cholecystokinin receptor (CCK-R) in the gallbladder.</p><p><b>METHODS</b>One hundred Guinea pigs were randomly divided into four groups, 25 animals for each. The control group was fed a standard diet, and the cholesterol group fed a diet containing 2% cholesterol. After taking the 2% cholesterol diet for two weeks, the natural group persisted on the standard diet, and the treated group was perfused by traditional Chinese medicine. Serum cholecystokinin (CCK) level in the portal vein and maximal binding capacity (B(max)) and Kd of CCK-R in the gallbladder were measured in the four groups by RIA and RBA, and the concentrations of cholesterol in bile were also observed.</p><p><b>RESULTS</b>Compared with the control group, after high-cholesterol feeding for two weeks, the gallbladder emptying rate [(65.83 +/- 7.32)% approximately (47.22 +/- 5.24)%] and B(max) of CCK-R [(60 +/- 27) approximately (32 +/- 13) fmol/mg protein] and in decreased fasting gallbladder volume (FV) [(0.89 +/- 0.26) approximately (1.34 +/- 0.61) cm(3)] and concentration of cholesterol [(0.44 +/- 0.11) approximately (0.60 +/- 0.13) mmol/L] in bile increased, but no change was in the serum CCK level and Kd of CCK-R in the cholesterol group. Compared with the natural group, after two-week in take of herb decoction of qingre lidan and liqi huoxue, FV [(1.27 +/- 0.60) approximately (0.90 +/- 0.27) cm(3)], RV [(0.85 +/- 0.45) approximately (0.32 +/- 0.12) cm(3)], FB [(0.92 +/- 0.35) approximately (0.73 +/- 0.21) cm(3)], RB [(0.76 +/- 0.34) approximately (0.29 +/- 0.08) cm(3)] in the treated group decreased significantly; but gallbladder emptying rate [(43.06 +/- 4.27)% approximately (67.01 +/- 6.82)%] increased significantly. The concentration of cholesterol in bile was lower in the treated group than in the natural group [(0.59 +/- 0.14) approximately (0.43 +/- 0.10) mmol/L], but no change was found in the serum CCK level. Bmax of CCK-R in the treated group increased significantly [(39 +/- 19) approximately (59 +/- 11) fmol/mg protein], Kd of CCK-R showed no significant changes between the treated group and natural group.</p><p><b>CONCLUSION</b>High cholesterol in gallbladder bile causes defective muscle contraction by down-regulating CCK-R in the gallbladder, so the reduction of cholesterol concentration of bile may contribute to gallbladder contraction.</p>

ABSTRACT

Objective To study the relationship between the expression of vascular endothelial growth factor (VEGF) and invasion, metastasis of primary hepatocellular carcinoma (PHCC). Methods Paraffin-embedded specimens from 30 patients with PHCC undergoing radical resection were studied. Agiogenesis was assessed by the expression of VEGF and microvascular density (MVD) using LSAB immunohistochemical staining. Results (1) The level of VEGF and MVD in  PHCC was higher than that in the paratumorous tissue (P<0.05). (2) The level of VEGF expression and MVD in PHCC with tumor metastasis or in nonencapsulated tumors were higher than that in PHCC without metastasis or in well encapsulated tumors (P<0.05).  (3) There was no significant differance in the expression of VEGF and MVD between large PHCC (diameter >5cm) and small ones (diameter ≤5cm). (4) The expression of VEGF was positively correlated with MVD (P<0.01).  Conclusions The expression of VEGF and MVD are highly correlated with invasion and metastasis in PHCC, may serve as a parameter for determining tumor metastasis and prognosis which may suggest a new idea to supervise and treat PHCC.

ABSTRACT

Objective:To study the effect of growth hormone(GH) on acute lung injury induced by endotoximia and its’ mechanism.Methods:After introduction of acute lung injury(ALI) by endotoximia,one hundred and tweleve male Sprague-Dawley rats were randomly divided into ALI group and GH group. Western blot,immunofluorescence staining and semi-quantitive reverse transcription polymerase chain reaction(RT-PCR) were used to determine the expression and activation of nuclear factor kappa B(NF-?B), the levels of CC16 protein and the expression levels of CC16 mRNA in ALI rats’ lung.Results:Both the expression levels of CC16 mRNA and the contents of CC16 protein in ALI rats’ lungs began to decrease significantly 0.5 h post-injury,reached nadir at 6 h post-injury,and then began to recover.Both the expression levels of CC16 mRNA and the contents of CC16 protein in GH group were significantly lower than those in ALI group at different time intervals post-injury.The dynamic changes of the expression and activation of NF-?B were contrast to those of CC16 protein levels.Correlation analysis indicated that CC16 correlates significantly with the extent of lung injury and the expression and activation of NF-?B.Conclusion:Down-regulation of CC16 expression plays a critical role in the pathogenesis of acute lung injury induced by endotoximia.The application of GH can deteriorate the lung injury induced by endotoximia through down-regulating the expression of CC16.
